A novel method for creating circular antisense oligonucleotides (circular ASOs) with enhanced stability and nuclease resistance for gene therapy is proposed. The closed-loop structure provides exonuclease resistance and an extended half-life in vivo. A double-crossover (DX tile) assembly method was used to bring ASO ends together for chemical ligation. Successful circularization was confirmed by gel electrophoresis. Circular ASOs show promise for gene knockdown. Future research will focus on their delivery to cells and in vitro efficacy.
