This study aims to engineer Escherichia coli for cellulose-based isobutene production. A heterologous cellulase (Cel-Tfu) will be designed for extracellular cellulose hydrolysis, generating glucose as a carbon source. Through metabolic engineering, a codon-optimized α-ketoisocaproate dioxygenase (KICD) from Rattus norvegicus will convert α-ketoisocaproate into isobutene, which will be quantified using gas chromatography. This research establishes a synthetic pathway for sustainable isobutene production.
